Theriva Biologics (AMEX: TOVX) is advancing its oncolytic virus therapy, VCN-01, which targets the dense stroma surrounding solid tumors to improve treatment efficacy. The Rockville, Maryland-based company reported encouraging results from clinical trials for pancreatic cancer (PDAC), retinoblastoma, and head and neck squamous cell carcinomas (HNSCC). VCN-01 is designed to degrade the extracellular matrix that often limits chemotherapy penetration and suppresses immune response, potentially enhancing the effectiveness of co-administered therapies.
The company is conducting trials in collaboration with the School of Medicine at the University of Leeds and the University of Pennsylvania's Perelman School of Medicine. These partnerships aim to evaluate VCN-01 across multiple hard-to-treat cancers, addressing a global solid tumor treatment market projected to reach $532 billion by 2032, according to Precedence Research.
Theriva is also developing next-generation oncolytic viruses, such as VCN-11, which incorporates novel Albumin Shield technology. This work is carried out with researchers from the Institut d'Investigació Biomèdica de Bellvitge (IDIBELL) and the Catalan Institute of Oncology (ICO). Additionally, the company is exploring treatments for hematologic cancers, including an approach to prevent acute graft-versus-host disease (aGVHD) in partnership with Washington University in St. Louis.
The potential impact of Theriva's work extends beyond individual patient outcomes. The global retinoblastoma treatment market alone is expected to exceed $3.3 billion by 2030, according to Grand View Research. Dr. Rocio Garcia-Carbonero, principal investigator for Theriva's main clinical site in Spain, noted that VCN-01's unique mechanism induces a robust immune response against cancer, setting it apart from traditional chemotherapy.
